Principle

a. Identification test
In this experiment, compound present in the Senna leaf is examine by using
thin-layer chromatographic. Different solvent with different polarity was
mixed and used as a mobile phase to get good separation. Plate was sprayed
with potassium hydroxide R in alcohol (50 % V/V) R for visualization.
b. Determination of Anthraquinone
Mixture of powdered drug and water were heated to separate the volatile
compound. The mixture is heated in water bath to prevent direct heating and
overheating. Heating process was carried out under reflux to return the heavier
components of the vapor to the boiling liquid. Chloroform is added to the solution to separate the
chlorophyll while addition of sodium hydrogen carbonate is to neutralize acid in the solution.
