Abstract
AllmajorABObloodallelesarefoundinmostpopulationsworldwide,whereasthemajorityofNativeAmericansarenearly
exclusivelyintheOgroup.OallelemolecularcharacterizationcouldaidinelucidatingthepossiblecausesofgroupOpredominance
inNativeAmericanpopulations.Inthiswork,westudiedexon6and7sequencediversityin180Obloodgroupindividualsfromfour
differentMesoamericanpopulations.Additionally,acomparativeanalysisofgeneticdiversityandpopulationstructureincludingSouth
Americanpopulationswasperformed.ResultsrevealednosignificantdifferencesamongMesoamericanandSouthAmericangroups,
butshowedsignificantdifferenceswithinpopulationgroupsattributabletopreviouslydetecteddifferencesingeneticdriftandfounder
effectsthroughouttheAmericancontinent.Interestingly,inallAmericanpopulations,thesamesetofhaplotypesO(1),O(1v),and
O(1v(G542A))waspresent,suggestingthefollowing:(1)thattheyconstitutethemaingeneticpoolofthefoundingpopulationofthe
Americasand(2)thattheyderivefromthesameancestralsource,partiallysupportingthesinglefoundingpopulationhypothesis.In
addition,theconsistentandrestrictedpresenceoftheG542AmutationinNativeAmericanscomparedtoworldwidepopulations
allowsittobeemployedasanAncestryinformativemarker(AIM).PresentknowledgeofthepeoplingoftheAmericasallowsthe
predictionofthewayinwhichtheG542AmutationcouldhaveemergedinBeringia,probablyduringthedifferentiationprocessof
Asianlineagesthatgaverisetothefoundingpopulationofthecontinent.
