CITRATE UTILIZATION TEST

AIM:
To demonstrate the ability of an organism to utilize citrate as sole source of
carbon.
PRINCIPLE:
Certain organisms can utilize citrate as sole carbon source and grow. During the
growth, acetate and other alkaline carbonates are produced. This reaction is shown by the
change in the color of the indicator.
MATERIALS REQUIRED:

6-8 hours old bacterial culture.

Simmon Citrate agar.

Inoculation loop.

Spirit lamp.

Test tubes.

PROCEDURE:
a. Simmon Citrate agar was prepared, sterilized and the slants were made in the test
tubes.
b. The cultures were then inoculated using inoculating loops.
c. After inoculation, the tubes were incubated at 35+2C for 24-48 hours.
