Purpose: To ensure the proper handling of Refractometer. Scope : The SOP is applicable for the handling of Refractometer. Responsibility HOD Quality QC Chemist Definition: SOP: Standard Operating Procedure Procedure: Zero Adjustment (Calibration): The instrument is carefully adjusted, but it is well to verify the setting by applying the glass test piece to the prism using minimum quantity of contact liquid and nothing if the reading is that marked on the test piece. If this is not the case, adjust the small squared screw at the back of the telescope with the key provided until agreement is secured. The mean of several reading be taken. Check the adjustment with distilled water in the way used for measuring liquid in light transmitted at grazing incidence. The temperature coefficient of the Refractive index of liquid is large, so the precise temperature of the measuring prism is carefully noted. The table below gives the refractive index of distilled water at temperature between 15 and 30 oC. If the average of several careful determinations at a certain temperature differs from the value in the table by no more than two ten thousands of a unit, the adjustment may be considered sufficiently accurate. If the position of the boundary line differs only slightly from the theoretically value, there is no need to for any readjustment if the difference is estimated and used as a correction factor. Deg. Measurement of Refractive Index of Liquids: 1. Open the prism box and clean both the prism surface gently using clean linen moistened in alcohol. 2. Place a few drops of sample to be tested on the ground surface of a lower prism. 3. Close and fasten the prism box gently. 4. The liquid be squeezed into a thin film between the prism.

5. Use an artificial source of light place conveniently in front of the instrument so that the light may be reflected into the prism box with the mirror until the illumination is good. 6. Focus the crosswire of the eyepiece & adjust the mirror until the illumination is good. Gently turn the prism box by means of the milled head screw (Control Knob) toward or backward until a colored band appears in the field of view. 7. By turning the milled ring at the base of the telescope, a position is found at which the colored band is eliminated, the field of view being partly light and partly dark with a sharp dark line. 8. Bring this sharp dividing line into coincidence with the intersection of the crosswire and read the refractive index or the brix scale which coincides on the divided line by the means of the magnifier. 9. Intermediate values can be easily estimated. Measurement of Refractive Index of Solids: 1. For the examination of plastic bodies such as resin, remove the lower prism box, soften the material by heat, and a smear a little contact liquid on the exposed surface of the upper prism. 2. Use an artificial source of light place conveniently in front of the instrument so that the light may be reflected into the prism box with the mirror until the illumination is good. 3. Focus the crosswire of the eyepiece & adjust the mirror until the illumination is good.
