Product overview INTENDED USE

The Rat TNF alpha ELISA is to be used for the

in-vitro quantitative determination of rat Tumor
Necrosis Factor. The assay will recognize both
natural and recombinant Rat TNF alpha. This
kit has been configured for research use only
and is not to be used in diagnostic procedures.

PRINCIPLE OF THE METHOD

The rat TNF alpha Kit is a solid phase sandwich

Enzyme Linked-Immuno- Sorbent Assay
 (ELISA). A polyclonal antibody specific for rat
TNF alpha has been coated onto the wells of
the microtiter strips provided. Samples,
including standards of known rat TNF alpha
concentrations, and unknowns are pipetted
into these wells.
During the first incubation, the rat TNF alpha
antigen and a biotinylated polyclonal antibody
specific for rat TNF lpha are simultaneously
incubated.
After washing, the enzyme ( streptavidin-
peroxydase ) is added. After incubation and
washing to remove all the unbound enzyme, a
substrate solution which is acting on the bound
enzyme is added to induce a coloured reaction
product. The intensity of this coloured product
is directly proportional to the concentration of
rat TNF alpha present in the samples.

Relevance Tumor Necrosis Factor Alpha (TNF alpha) is a
protein secreted by lipopolysaccharide
stimulated macrophages, and causes tumor
necrosis when injected into tumour bearing
mice. TNF alpha is believed to mediate
pathogenic shock and tissue injury associated
with endotoxemia. TNF alpha exists as a
multimer of two, three, or five noncovalently
linked units, but shows a single 17 kDa band
following SDS PAGE under non reducing
conditions. TNF alpha is closely related to the
25 kDa protein Tumour Necrosis Factor beta
(lymphotoxin), sharing the same receptors and
cellular actions. TNF alpha causes cytolysis or
cytostasis of certain transformed cells, being
synergistic with interferon gamma in its
cytotoxicity. Although it has little effect on
many cultured normal human cells, TNF alpha
appears to be directly toxic to vascular
endothelial cells. Other actions of TNF alpha
include stimulating growth of human
fibroblasts and other cell lines, activating
polymorphonuclear neutrophils and
osteoclasts, and induction of interleukin 1,
prostaglandin E2 and collagenase production.
 TNF alpha is currently being evaluated in
treatment of certain cancers and AIDS Related
Complex.
