Title: Immunohistochemistry and Microscopy
Brief background on the CET/S:
The CvSU College of Veterinary Medicine and Biomedical Sciences (CVMBS) has
been working towards excellence in the field of veterinary education in the country. The
College has been awarded Level 2 accreditation by the Accrediting Agency of Chartered
Colleges and Universities of the Philippines (AACCUP) for the period of December 16,
2012 to December 15, 2016 and Center of Development (COD) in Veterinary Medicine by
the Commission of Higher Education (CHED) for the period of February 27, 2009 to
February 27, 2012. With these developments, the College has become more committed to
respond to the ever changing needs of the country, particularly in field of veterinary
education and research.
In 2009, the College has acquired equipment to establish an Immunohistochemistry
(IHC) and Microscopy Laboratory that will cater to veterinary researches, as well as
biomedical studies. Unfortunately, the personnel previously trained for this type of
laboratory is no longer connected with the University. Continuous technical trainings on
immunostaining and microscopy were needed to fully operationalize the laboratory which
is why the College conducted a short course on Immunohistochemistry for the Faculty and
staff of the which was facilitated by experts from the College of Veterinary Medicine at
University of the Philippines, Los Baos. And just recently, a faculty member attended an
Immunohistochemistry and Microscopy Course at the Marine Biological Laboratory at
Woods Hole, Massachusetts, USA. With these, the utilization of the laboratory for
research, and even for instruction, may be maximized.
To date, the laboratory has equipment which approximately costs around
7,000,000. These include: (1) automated rotary tissue processor; (2) tissue embedding
console, (3) rotary microtome, (4) Nikon Eclipse 90i state-of-the-art microscope for bright
field and fluorescence microscopy, (4) immunohistochemistry station, (5) ultra low
biological freezer, and (6) ELISA reader. However, to further enhance the capability and
proficiency of the laboratory in the application of the IHC technique to a broader scope,
additional equipment, materials and reagents are needed to be purchased.
Immunohistochemistry or IHC is widely used in basic research to understand the
distribution and localization of biomarkers and differentially expressed proteins in different
parts of a biological tissue using the process of detecting antigens (e.g., proteins) in cells

of a tissue section by exploiting the principle of antibodies binding specifically to antigens.


Clinically, immunohistochemical staining is widely used in the diagnosis of abnormal cells
such as those found in cancerous tumors and maybe used to detect antigens of
pathogens in tissue such as rabies in the brain (IFAT). IHC has certainly evolved over the
last few years to become an invaluable technique in many facets of life sciences research
including veterinary medicine.
When using IHC to understand the spatial location of function of a molecule in
tissue, it is important to be confident that the antibodies used bind specifically to their
target epitope. When antibodies are not validated or when IHC protocols are not
optimized, non-specific binding will result in incorrect findings and conclusions. Therefore,
when using this technique, it is critical to take the time to properly validate the specificity of
an antibody and to optimize the protocol in performing a study. When done properly, one
can be confident that the IHC performed is providing the correct information about
changes to the molecule of interest in the tissue.
Western blots, pulse field gel electrophoresis and ELISA are assays similar to IHC
in the sense that they utilize antibody specificity and quantitative changes in the
expression of proteins either in individual cells or biological samples such as tissue
homogenates. If the findings from studies these techniques are in agreement with the
findings of IHC, they allow the investigator to be more confident that the results obtained
are correct.
Similarly, the collection and measurement of mRNA in cells or tissue homogenates
using real-time quantitative polymerase chain reaction (RT-PCR) can confirm findings with
IHC. Effecting similar changes in the mRNA for the target epitopes as measured with IHC
and quantitative microscopy augment the confirmation of the specificity of an antibody.
The cutting edge technology proposal is geared toward building the proficiency of
the IHC laboratory as a tool for veterinary research and diagnosis. It has three
components: (1) basic research, (2) training, and (3) technology development.
Moreover, the IHC technique may not only be used in veterinary research but will
also cater to the research needs of the Cavite State University as the National Coffee
Center of the Philippines. There is growing popularity of Civet Coffee or Kape Alamid in
the Philippines. Researches on Civet Coffee production may boost the profitability and
sustainability of enterprise. Furthermore, it will serve as a boost for the sleeping coffee
industry creating more employment and business opportunities.

Executive Summary
Summary of Cutting-Edge Technology
IMMUNOHISTOCHEMISTRY AND MICROSCOPY PROGRAM

INTRODUCTION
In 2009, the College has acquired equipment to establish an Immunohistochemistry
(IHC) and Microscopy Laboratory that will cater to researches foremost, in the veterinary
field. Continuing technical trainings on immunostaining and microscopy is needed to fully
operationalize

the

laboratory.

Just

recently,

faculty

member

attended

an

Immunohistochemistry and Microscopy Course at the Marine Biological Laboratory at


Woods Hole, Massachusetts, USA. With this development, it is envisioned by the College
to use the expertise learned from the training to maximize the potential of the laboratory
for research. To date, the laboratory has the following equipments: (1) automated rotary
tissue processor; (2) tissue embedding console, (3) rotary microtome, (4) Nikon Eclipse
90i state-of-the-art microscope for bright field and fluorescence microscopy, (4)
immunohistochemistry station, (5) ultra low biological freezer, (6) ELISA reader. However,
there is a need to develop the proficiency of the laboratory in actual running of the IHC
technique. To do this, additional equipments and reagents are needed to be purchased to
maximize the potential of the laboratory. Equipments which incudes (1) RT-PCR, (2) Pulse
Field Gel Electrophoresis and (3) Western blot will be utilized to build proficiency in the
IHC technique.
The cutting edge program has three components: (1) basic research, (2) training
and (3) technology development.
A. Basic Research
Project Title: Immunohistochemistry of the Pancreatic Islets of Selected Wildlife in the
Philippines
Study 1 Title: Immunohistochemistry of the Pancreatic Islets of Selected
Frugivorous and Insectivorous Bats in the Philippines (Order
Chiroptera)
Study 2. Title: Immunohistochemistry of the Pancreatic Islets of Frugivorous and
Carnivorous Monitor Lizard in the Philippines (Genus Varanus)

Objectives:
The general objective of the study is to document, describe and differentiate histological
characteristics of the endocrine pancreas of certain wildlife species using
immunohistochemistry. Specifically, this study will be conducted to identify insulin and
glucagon reactive pancreatic cells and to compare and contrast the pancreatic islet size,
density, location of islet cells of selected species. This study shall investigate the
morphological adaptation of the endocrine pancreas toward diet preferences of some
Philippine wildlife. The description of the morphophysiological features can be used to
deduce ecological and evolutionary implication of body forms.
Expected outputs
The study is expected to provide data on the morphophysiological properties of tissues
from various endemic wildlife. The study is also expected to help increase the proficiency
of CVMBS personnel in doing immunohistochemistry work for research and instruction
purposes. The study also will also give them the chance to publish results the field of
immunohistochemistry and microscopy in prestigious journals and periodicals.
Target beneficiaries

seeks more research endeavors from its constituents to attain global recognition

students and professional alike.

Immunohistochemistry
Objectives
1. Augment the diagnosis, treatment and control of economically important animal
disease
2. Increase productivity of livestock farmers by definitive diagnosis of animal diseases
using IHC technique
3. Augment the diagnosis, treatment and control of zoonotic diseases

To generate scientific data on diseases affecting livestock which have economic and
public health importance

Upgrading of Facilities
Rationale
Improvement of the colleges laboratories shall strengthen instruction by
providing hands-on experience of students and shall enhance its capability to
conduct researches as well as provide diagnostic services to its clientele. In addition,
purchased equipments shall complement evaluation and validation of IHC technique
in the innovative research project of this proposal.

Activities (narrative explanation of activities)


a. Purchase equipments for the evaluation and validation of IHC technique in the
innovative research project of this proposal
RT-PCR, Pulse Field Gel Electrophoresis and Western Blot serves as a
tool to evaluate and validate the IHC technique/protocol in the innovative
research project of this proposal
b. Purchase of state-of-the-art equipments for the Upgrade of Animal Disease
Diagnostic Laboratory (ADDL)
ADDL serves as tool for diagnosis. Upgrading includes acquisition of
basic equipments for evaluation blood chemistry profiles of animal patients and
animals of concern for research. RT-PCR and Pulse Field Gel Electrophoresis
are also used as a diagnostic tool for animal diseases.
c. Purchase of equipment for Enhancing the Diagnostic Imaging Laboratory
Additional purchase of rectal ultrasonograph probe shall enhance the
capability of the existing ultrasound machine for obstetrical and
theriogenological purposes.
d. Purchase of Biological Safety Cabinet
Handling of certain materials for research and diagnostic may pose
occupational and environmental hazard. Purchase of a biological safety cabinet
will minimize such dangers.

2. Research Capability BuildingHuman Resources and Facilities (Technology Development)


Technology Development for Animal Disease Diagnosis using IHC needs purchase of
other reagents, materials, and miscellaneous equipments (i.e liquid nitrogen storage tank,
pippetors). Likewise, upgrades of existing state-of-the-art microscope (i.e. microscope
objectives and excitation filters), purchase of latest specification computers and quantitative
microscopy softwares are needed to support the research capability for the technology
development component of this proposal.

Research generated will be presented in various fora. Funding support for paper
presentation will encourage researchers involved in the project to present their works in
various scientific fora.
6. Local and National Conferences
Continuing education and establishment of future collaboration are some of the
outcomes of attending local and national conferences. Funding support will encourage faculty
members to attend relevant conferences to gain up-to-date information and establish
partnerships for research and development activities.
7. Exhibits/fairs, IEC and Commercialization
IEC materials and activities with regards to IHC and microscopy is important in the
promotion and dissemination of the technology.
8. Industry Government - Academic Linkage Projects
Establish linkages to industry for support for the innovative research project.
Partnership with industry stakeholders may be a possible source of extramural funding for the
sustained development and utilization of the technology.
