Tissuecultureisthegrowthoftissuesorcellsseparatefromtheorganism.
Thisistypicallyfacilitatedviauseofaliquid,semisolid,orsolidgrowth
medium,suchasbrothoragar.Tissueculturecommonlyreferstothe
cultureofanimalcellsandtissues,withthemorespecifictermplanttissue
culturebeingusedforplants.Theterm"tissueculture"wascoinedby
AmericanpathologistMontroseThomasBurrows,M.D.[1]

In1885WilhelmRouxremovedasectionofthemedullaryplateofan
embryonicchickenandmaintaineditinawarmsalinesolutionforseveral
days,establishingthebasicprincipleoftissueculture.In1907thezoologistRossGranvilleHarrisondemonstrated
thegrowthoffrogembryoniccellsthatwouldgiverisetonervecellsinamediumofclottedlymph.In1913,E.
Steinhardt,C.Israeli,andR.A.Lambertgrewvacciniavirusinfragmentsofguineapigcornealtissue.[2]In1996,
thefirstuseofregenerativetissuewasusedtoreplaceasmalllengthofurethra,whichledtotheunderstandingthat
thetechniqueofobtainingsamplesoftissue,growingitoutsidethebodywithoutascaffold,andreapplyingit,can
beusedforonlysmalldistancesoflessthan1cm.[3]
GottliebHaberlandtfirstpointedoutthepossibilitiesofthecultureofisolatedtissues,planttissueculture.[4]He
suggestedthatthepotentialitiesofindividualcellsviatissuecultureaswellasthatthereciprocalinfluencesof
tissuesononeanothercouldbedeterminedbythismethod.SinceHaberlandt'soriginalassertions,methodsfor
tissueandcellculturehavebeenrealized,leadingtosignificantdiscoveriesinbiologyandmedicine.Hisoriginal
idea,presentedin1902,wascalledtotipotentiality:Theoreticallyallplantcellsareabletogiverisetoacomplete
plant.[5][6][7]

Modernusage
Inmodernusage,tissueculturegenerallyreferstothegrowthofcellsfromatissuefromamulticellularorganism
invitro.Thesecellsmaybecellsisolatedfromadonororganism,"primarycells",oranimmortalisedcellline.The
cellsarebathedinaculturemedium,whichcontainsessentialnutrientsandenergysourcesnecessaryforthecells'
survival.[8]Thetermtissuecultureisoftenusedinterchangeablywithcellculture
Theliteralmeaningoftissueculturereferstotheculturingoftissuepieces,i.e.explantculture.
