1. OBJECTIVE

Objective of this SOP is to define a sampling procedure for purified water and drinking water
as well.

2. SCOPE

This SOP describes the execution of purified and drinking water sampling.

3. RESPONSIBILITIES

4.1. Microbiologist:

- To interface this SOP to the actual practices of flow patterns.

- To ensure that the SOP is updated & reflects the actual execution.

- To review the SOP in case of any change or whenever it is due for revision.

4.2. AM Microbiology:

- To follow the instructions laid down by this SOP.

- To keep the relevant SOPs valid and updated.

4. REQUIREMENTS
4.1. Refrigerator
4.2. Sterile screw-capped glass bottles 100 mL
4.3. Sterile latex gloves
4.4. Filtered 70% Isopropyl Alcohol spray bottle
4.5. Washed and cleaned 2.5 L bottle

5. SAMPLING PROCEDURE FOR MICROBIOLOGICAL ANALYSIS


5.1. Sampling points must be labeled properly as no any difficulty to take
representative sample.

5.2. Before sampling, properly label the sampling bottle including sampling
point number, sampling date and name.

5.3. Sampling points must be free from aerators, sieves and purification
devices.

5.4. Before sampling, wash hands thoroughly, do not open sterile sample
bottle until you are ready to proceed.

5.5. Wear sterile latex gloves; disinfect them with filtered 70% I.P.A.

5.6. Before sampling, remove the aluminium cover from the sampling point
and disinfect it with filtered 70% I.P.A. for removing viable as well as non-
viable particles from the site of sampling, if any.

5.7. Open the sampling point and drain-out the water approximately for
1 minute (about 2 Liters); ensure that there is no splattering in water flow.

5.8. Take sterile sample bottle near to the sampling point, hold it from the
bottom in left hand and with right hand slowly remove the cap, make sure

that the inner surface of cap should not come in contact with any surface
including fingers or hands of the individual collecting the sample.

5.9. Collect the water sample in the sterile sample bottle up to 100 mL (do not
overflow the sample bottle) from mid of the water stream.

5.10. During collection of the sample, do not allow the bottle or the water
sample in the bottle to come in contact with the sampling valve.

5.11. Place the cap on the bottle as quickly as possible without heavy
movement of sample towards cap (avoid air to trap inside the sample
bottle).

5.12. Tighten the cap securely, close the sampling valve.

5.13. Collect the sample(s) and proceed for analysis as per SOP # .
If the sample(s) are not taken immediately for analysis OR not analyze
within 30 minutes to 2 hours then store at 2 8 C in refrigerator but not
exceed 24 hours.

5.14. Ensure that the analysis is, in any case, initiated within 24 hours after
sampling.

6. SAMPLING PROCEDURE FOR CHEMICAL ANALYSIS


6.1. For chemical analysis, clean and dried sample bottles are used for
sampling.

6.2. Before sampling, properly label the sampling bottle including sampling
point number, sampling date and name.

6.3. Before sampling, wear gloves and remove the aluminium cover from the
sampling point.

6.4. Open the sampling point and drain-out the water approximately for
1 minute (about 2 Liters); ensure that there is no splattering in water flow.

6.5. Take sample bottle near to the sampling point, hold it from the bottom in
left hand and with right hand slowly remove the cap, make sure that the

6.6. Rinse the bottle with the water to be sampled at least 3 times.

6.7. Collect the water sample in the sample bottle up to the mark, place cap
on the bottle as quickly as possible and ensure that no air bubble is
entrapped.

6.8. For TOC analysis, collect the sample in the separate bottle.

6.9. Collect the sample by over flowing the bottle and close with the cap
immediately.

6.10. Tighten the cap securely, close the sampling valve.

6.11. Collect the sample(s) and proceed for analysis as per SOP # .
If the sample(s) are not taken immediately for analysis OR not analyze
within 30 minutes to 2 hours then store at 2 8 C in refrigerator but not
exceed 24 hours.

6.12. Ensure that the analysis is, in any case, initiated within 24 hours after
sampling.

7. PRECAUTIONS

7.1. Always wear latex gloves either sampling for microbiological analysis OR
chemical analysis.

7.2. Ensure that there is no splattering OR any other turbulence in water


stream.

7.3. Avoid heavy movement of sample bottle (microbiological analysis) while it


is opened.

7.4. Do not touch inner surface of cap and also care must be taken during
sampling that nothing come in contact with sampling valve either it is
bottle OR water that is being sampled.

7.5. Water sample analysis should be initiated immediately as sample


received otherwise sample should be kept in refrigerator at 2 8 C but
not more than 24 hours.

8. TRAINING

Training about the contents of this SOP must be imparted to all the concerned
personnel once in a year and also to new employees before starting with the job and
the records documented according to training SOP.
