EXPERIMENT 3 : ESTERIFICATION REACTIONS OF VANILIN : THE USE OF NMR TO DETERMINE A
STRUCTURE.

OBJECTIVES:

1. To purpose mechanism that will explain why the reaction proceeds differently under acidic
and basic conditions.
2. To investigate the reactions between vanilin and acetic anhydride in two conditions, basic
and acidic condition.
3. To identify the product formed in both acid and base reactions.
4. To obtain IR spectrum and melting points of the products.

INTRODUCTION:

PROCEDURE:

A. Preparation of 4-acetoxy-3-Metloxybenzaldehyde (Vanilin Acetate)

1. In 250 ml Erlemeyer flask, 1.50g vanilin is dissolved in 25 ml of 10% sodium hydroxide


(NaOH).
2. 30g of crushed ice and 4.0 ml of acetic anhydride are added to the flask.
3. The flask is stopped with clean rubber stopper and it is shaked several times over a 20
minutes (a cloudy, milk white precipitate is formed immediately while adding the acetic
anhydride).
4. The precipitated is filtered using Buncher funnel and the solid is washed with three portions
of 5 ml ice cold water.
5. The solid recrystallized from 95% ethyl alcohol.
6. The mixture is heated in a hot water bath at about 60 ᵒc to avoid melting the solid.
7. When the crystal is dried, it is weighed and percentage yield is calculated.
8. The IR spectrum of product is obtained by using dry film method.
