2. Preface
An object that will be seen by a microscope is called microslide or
preparations. Prepare the preparation before observing an object
by microscope. Solid preparation such as animal and plant tissues
can be made by slicing a thin tissue section transversely or
longitudinally, then put it on an object glass and covered it with a
cover glass. To make the part of the cell tissues clearly visible, the
object is given a dye solution or iodine. Preparations of liquid
materials such as microorganisms in water and liquid substances
can be made by the dripping the fluid on an object glass slide and
cover it with a cover glass.

BEM 126 Multimedia Microscope | 33

3. Procedure
1. Make preparation of dry material.
a. Clean the object glass and the cover with cloth or tissue.
It should be dirt or dust free.
b. Slice the leaf with knife or cutter in cross section.
Note: The slice result should be thin and upright.
c. Place the slice on the object glass that have been given a
drop of water, using tweezers or a toothpick.

d. Slowly cover the slide with cover glass. One side of the
cover glass should touch the edge of water drop in a tilt
o
position of about 45 angle.
e. Excess water can be removed by placing filter paper or
tissue in the edge of the cover glass.
f. Preparation is ready to be observed under the
microscope.
2. Make preparation of liquid material.
a. Clean the object glass and its cover with cloth or tissue. It
should be dirt or dust free.
b. Place one drop of pond water on the object glass by
using a pipette. Slowly cover it with a cover glass by one
side of the cover glass touch the edge of water drop in a
tilt position of about 45° angle. (Look at the picture above
and ask the teacher to demonstrate it).
c. Excess water can be removed by placing the filter paper
or tissue in the edge of the cover glass.
d. Preparation is ready to be observed under the
microscope.
