Learning Target:
To identify tissue type of cheek tissue and observe mitosis in onion (Allium cepa) root tips and understand
mitotic cell division.
Materials:
Activity 1
Cotton swab
Slides
Cover Slip
Methylene Blue
Bunsen burner
Dropper
Procedure:
Activity 1
1. Take a cotton swab and rub the inside of your cheeks to collect cheek tissue sample.
2. Smear these cells on a microscopy slide
3. Completely air-dry the slide, without applying heat.
4. Heat fix the dried slide by quickly pulling it through a Bunsen burner (2x), but in a way that the cells do not
touch the flame. Pull it through the flame with the cells on top and the flame below
5. Apply a drop of methylene blue to the heat fixed but cold specimen slide. Allow the stain to work for a few
minutes and then carefully rinse away the stain with water. Do not apply the water stream to the cells directly.
Allow the water to run over the cells from the top of the slide. Air dry the slide.
6. Observe with the microscope. Locate the tissue sample under LPO then switch to HPO.
Activity 2
1. Obtain a prepared slide of an onion root tip where cells were actively dividing.
2. Place the slide on the microscope stage and use the low power objective (LPO) to locate and focus on the root
tip until it is clearly visible and switch to high power objective (HPO).
3. Locate the cells that were in the stage of interphase, prophase, metaphase, anaphase, and telophase then draw
them in the boxes below.
