Manipulation of the Microscope

 To see image of object under microscope, objective must be adjusted at


proper distance from object
Focusing the Microscope
 Adjust the distance of the lenses to the object so that a clear image will be
seen though ocular
Directions:
1. Gently place Microscope and Position on table.
2. Align LPO with optical axis of microscope.
3. Open diaphragm and for disk type, use largest aperture (opening controlling
amount of light passing through).
4. Turn concave side of mirror towards light source and regulate diaphragm
until reflected light gives clear and even-lighted field.
5. Prepare specimen (Letter e).
6. Raise body tube through course adjustment knob (counterclockwise/towards
you) until LPO is 2cm away from stage surface. Place and secure slide.
7. Turn Coarse Adjustment Knob (clockwise/away from you) until LPO is
1mm away from slide. Never let cover slide and objective touch.
8. Using both eyes to look through ocular, turn coarse adjustment knob until
image comes into clear focus. Adjust position of letter until image appears.
Remember the working distance (distance between cover slip and objective).
9. Bring image into sharper focus using fine adjustment knob, slowly move
slide to the left then right.
10.Move slide and examine position of slide
11.Without altering adjustment, shift to HPO through revolving nosepiece. If
par-focal, enlarged but blurred image will be seen. Adjust with fine
adjustment knob until clear. Never use coarse adjustment knob when using
HPO.
12.Practice moving slide under different objectives until you can make slight
movement with ease (important in pointing particular parts with pointer).
13.Always examine under LPO first to get good general view of specimen. It is
more difficult to locate object with HPO and there is greater chance of
breakage. It can also cause eye strain, dizziness or headache.
