Observed how to use The Beckman Coulter


1 machine to load blood samples so that Complete
Complete Blood Count sample Loading Blood Count can be performed

Learned how to perform the ESR test.


Erythrocyte Sedimentation Rate (ESR) Westergren method is used, 2 ml of
venous blood from EDTA tube is added to
a tube containing 0 .5 ml of sodium
citrate. The blood is drawn into a
2 Westergren-Katz tube to the 200 mm
mark. The tube is placed in a rack in a
strictly vertical position for 1 hour at room
temperature, in which the red blood
cells will slowly settle at the bottom.
This will leave a clear, yellowish fluid
at the top, which is blood plasma.The
result of the test will depend on the
amount of plasma at the top of the
tube after 1 hour. The measurement
will be in millimeters per hour
(mm/hr). This test usually indicates
inflammation with no specific
diagnosis.

Observed and learned how to prepare a PBF by


adding one small drop of blood to one end and
Preparing a Peripheral Blood Film (PBF), then by using a another clean slide and hold it
staining to an angle of 45 deg touch the blood with one
end of the slide so the blood runs along the
edge of the slide. Allow it to dry then stain it by
the Romanowsky stains which are neutral
stains composed of a mixture of oxidized
3 methylene blue (azure) dyes and Eosin Y.
The azures are basic dyes that bind acid
nuclei and result in a blue to purple color.
The acid dye, eosin, is attracted to the
alkaline cytoplasm, producing red
coloration. Finally allow it to dry and view it
under Microscope

Learned and observed how to view and report


the stained blood smear slide under the
Microscopy of the Peripheral Blood Film microscope , how to differentiate between RBCs

WBCs and platelets, also how to report different
kinds of conditions like anaemia. Moreover we
did differential count of the WBCs.
