I. Modified TRUE OR FALSE.

Write TRUE if the statement is correct but if it’s false, change

the underlined word or group of words to make the whole statement true. (10pts)

1. 2N HCl acid is used to help extraction of fatty acids from glycerides, glycolipids and
phospholipids by breaking covalent and ionic bonds of lipids to protein and carbohydrates.
2. Celite is added to aid in filtering.
3. Anhydrous ether is used in fat extraction because presence of moisture can decrease the
efficiency of fat extraction
4. In Soxhlet method, the sample is submerged into a boiling solvent to extract the fat and sugar
while in soxtec method (Randall method) the condensed solvent drips to the sample to extract
the fat
5. After acid hydrolysis, the sample tube is dried in the oven at 60 degrees Kelvin.
6. Presence of peroxides in anhydrous ether can cause an explosion.
7. During solvent extraction, the sample undergoes three processes namely extraction, rinsing and
drying.
8. The appropriate fat extraction method for samples with unbound fat such as butter is acid
hydrolysis.
9. During extraction , 25 cycles of solvent is needed to extract the fat in the sample
10. Peroxides can be detected by mixing ether with potassium nitrate.

II. Flow Chart or Schematic Diagram of Total Fat Analysis (10pts)

III. Computation (10pts)

A sample was submitted for proximate analysis. During sample preparation, the analyst added 80ml
deionized water to 125g sample before homogenizing the sample. Then the analyst weighed 10.0564g
sample for fat analysis.

a) Compute for fat content if the weight of fat extracted is 3.2564 g.

IV. Essay (5pts each)

a) Differentiate Soxhlet vs Soxtec method (Randall Method)? Explain the principle.

b) Why is anhydrous ether used in extraction compared to other solvent?
c) Why do other samples such as processed food need acid hydrolysis to extract total fat?
