Solution
Description
EcoDye™ DNA Staining Solution is an stable and sensitive DNA staining dye. This fluorescent dye can
be used on a agarose gel electrophoresis to replace Ethidium Bromide(EtBr) which has toxicity.
EcoDye™ DNA Staining Solution is a product which is non-mutagenic, non cytotoxic and it can be used
with mixing a small amount of it when making the agarose gel. EcoDye™ DNA Staining Solution can be
formed of a complex through strong binding with DNA and it can be detected through UV imaging or
LED system.

Protocol
① Add agarose powder (0.8 ~ 3 %) into 100 ㎖ of 1.0X TAE buffer and heat it thoroughly using
microwave.
② Add 10 ㎕ of EcoDye™ DNA Staining Solution and mix the solution avoid forming bubbles. After that,
pour it into the gel tray (Please use 5 ~ 10 ㎕ EcoDye™ DNA Staining Solution according to the
condition of experimental condition and gene.)
③ Allow the agarose gel to cool until solidified.
④ Load the sample DNA in the agarose gel after mixing with loading dye and perform electrophoresis.
(Please use 1.0X TAE buffer as a electrophoresis buffer.)
⑤ Detect the bands under UV or LED illuminator after electrophoresis step.

※ Note
Please wear gloves and goggles while handling. EcoDye™ DNA Staining Solution may irritate skin and eyes
although it has no toxicity.

Application
-In case of ratio of EcoDye™ DNA Staining Solution is lower than the recommended amount, it can affect
the movement of DNA fragment.
- The thickness of gel should be less than 0.5 cm since thick gels may decrease sensitivity.
- Adjust the amount of EcoDye™ DNA Staining Solution in case of making higher percentage of
agarose gel (Increase the volume of EcoDye ™ in case of that the percentage of agarose gel getting higher).
- Repeated melting of gels containing EcoDye™ may result in low sensitivity.
- Detect the bands under UV illuminator as quickly as possible. DNA can be damaged in case of
exposure to UV light for a long time.
- Store the EcoDye ™ DNA Staining Solution at the amber tube. The fluorescent dye can be degraded
by light.
- In case of storage for EcoDye ™ in agarose gel, please store it in the container which is to avoid the
light and recommended to use it within 2 weeks.

Check details when using EcoDye™ DNA Staining Solution
- Check of usage : Use EcoDye™ 10㎕ per 100㎖ of agarose solution.
- Check the thickness of agarose gel : The thickness of gel should be less than 0.5 cm.
- Adjust and use TAE buffer after dilution that the final concentration of TAE buffer could be 1.0X.

Recommended to increase the volume of EcoDye™ DNA Staining Solution in case of that
the percentage of agarose gel getting higher.

03. Check whether to exposure by light


Recommended to store the EcoDye ™ DNA Staining Solution at the amber tube because
the fluorescent dye can be degraded by light.

06. Check the expiration date
Expiration date is two years in case of storage at 4°C.
07. Check the loading condition
Recommended to load with sedimentation materials such as 6X loading dye to prevent the floating
of DNA and to check the movement of DNA visually.
08. Check the power supply and electrophoresis tank
- Check the power and fuse of power supply - Check the platinum in the electrophoresis tank
- Check if air bubbles is formed or not in the platinum of electrophoresis
09. Check the running time of electrophoresis
In case of prolonged running for electrophoresis, detection of the bands may not be
possible because DNA fragment could be fell out of agarose gel.
10. Check the UV lamp
In case of that UV lamp is not working properly, the bands could be showed weakly.
11. Check the grade of agarose gel
According to the grade of agarose, separated fragment size of DNA, movement speed of
DNA and the intensity of gel could be different. Recommended to choose the agarose
powder according to the experimental purpose.
12. TAE buffer check
Check the final concentration of electrophoresis buffer (TAE or TBE) and compatibility of
the buffer for electrophoresis according to the buffer type.

01. Check the final concentration of TAE buffer


In case that final concentration of TAE buffer is less than 0.5X, movement speed of the band can be
affected due to lack of ion in the buffer. Use final concentration of TAE buffer to over 0.5X.
*Use the TAE buffer of 1.0 X final concentration in case of electrophoresis for the sample which has
multiple target bands such as multiplex PCR products.

02. Check the percentage of agarose gel


In case of electrophoresis for multi band PCR products or small sized fragment, recommended to
electrophoresis using high concentrated agarose gel (over 2-3%).
In case of electrophoresis for large sized fragment, recommended to electrophoresis using low
concentrated agarose gel (less than 1%).
03. Check the running voltage of electrophoresis
In case of multi band PCR product, bands may not be separated at the high voltage.
Curved band -Check whether well is flat or not in the process of inserted comb.
