Kato thick Smear (Cellophane thick smear)

The cellophane-covered thick smear was originally detected for use in field surveys for helminth infections. The
technique is useful for helminth eggs with thock shells such as Ascaris and Trichuris but not for helminth larvae or
protozoa.
Procedure:
1. Label slide properly and place approximately 50-60 mg of stool (the size of a soy bean) at the center of a glass slide
and cover with a square piece of pretreated cellophane.
2. With the aid of a rubber stopper, press the cellophane gently to spread the stool specimen evenly to about the
circumference of the stopper. Make sure that the specimen will not spread beyond the cellophane cover.
3. Leave the prepared slide at room temperature for 10-20 minutes. During this time, the microscopic field becomes
clear due to the action of glycerine on stool sample.
4. Examine the preparation under the microscope after standing and look for helminth parasites under LPO to HPO.
Don't allow the slides to stand for long period (more than an hour) as it causes drying and shells of hookworm will
dissolve in glycerine solution.
5. Report results as:
Number of eggs of parasite name per smear
No ova seen - if negative for parasites

Kato-Katz Technique
This technique is useful for assessing the intensity of infection in schistosomiasis and common soil-transmitted
helminthiasis like ascariasis, trichuriasis and hookworm infection. Consistency of the stool is the main determinnat of
the sensitivity of this technique. This technique can only be done on fresh stools and not in liquid-preserved samples.

Procedure:
1. Label slide and position the template at the center of the slide.
2. Place the fecal sample on the cover of the stool container. Press or strain the fecal sample with the wire net. Scrape
the feces which passed through the net then transfer them to the central hole of the card or plastic template lying over
a clean glass slide.
3. Fill up the hole, flatten the top level and lift the card or the template leaving the fixed amount of feces on the slide.
Put the pre-treated cellophane over the fecal sample.
4. With the aid of a rubber stopper, press the cellophane gently to spread the stool specimen evenly to about the
circumference of the stopper. Make sure that the specimen will not spread beyond the cellophane cover.
5. Leave the prepared slide at room temperature for 10-20 minutes. During this time, the microscopic field becomes
clear due to the action of glycerine on stool sample.
6. Examine the preparation under the microscope after standing and look for helminth parasites under LPO to HPO.

7. Report the presence of parasites using this formula:
# eggs of parasite/gm feces = # of eggs of parasite counter X 1000 mg
43 mg gm
Note: Kato-katz template is able to measure about 43 mg of stool sample; thus parasite count from this preparation
represents parasite load in 43 mg feces.
