Contexte scientifique
Nous proposons donc dans le cadre d’un post doctorat sur financement Ifremer :
-D’une part de tester l’utilisation de la technique de DHPLC sur des échantillons d’ADN issus
de travaux de cultures de communautés précédents (Doctorat de N. Byrne) pour lesquels
nous possédons les données moléculaires de diversité (clonage et séquençage et suivi
temporel par SSCP). Ce travail pourra aboutir à la rédaction d’une publication
méthodologique.
-Et d’appliquer l’utilisation de cette technique à de nouvelles cultures de communautés en
bioréacteur, notamment en conditions d’autotrophie sous différentes conditions
physicochimiques.
-D’autre part de mettre en route une culture visant à l’enrichissement des micro-organismes
Anammox détectés dans des échantillons de cheminées hydrothermales, et d’estimer
l’enrichissement des micro-organismes Anammox par DHPLC et grâce à des mesures
d’activité1.
Demande financière :
La somme demandée concerne l’achat : d’une colonne haut débit pour la séparation
des acides nucléiques (3000€), des tampons d’élutions (1000€), d’une lampe de détection,
de pré-filtres et seringues d’injection (1000€) indispensables au fonctionnement de la
DHPLC, soit un total de 5000€

Ces travaux seront réalisée en étroite collaboration avec le laboratoire « Institute of Wetland and Water Ecology » à Nijmegen,
aux Pays-Bas, laboratoire qui dispose d’une compétence avérée dans ce domaine.
