The microorganisms are grown into four types of culture which is broth
culture, slant culture, stab culture and plate culture. Before streak the aseptic
techniques is performed to avoid contamination. Separate nutrient agar plates
are being used to perform two type of streaking which is dilute streaking and
single streaking using the bacterial cultures given. As for nutrient slants and
gelatine the bacterial culture is being stabbed into a a single line by the
inoculating needle. Nutrients slant the streaking is performed in a straight line.
Then the nutrient gelatine is incubated at 20°C and the rest at 37°C .
The result will aid in explaining the principles behind the use of solid and
broth media for culturing and describing different colony morphologies. As for
Bacillus cereus there are no growth on the deep nutrient agar but the rest
there are some sign of formation of the bacteria culture. While
Staphylococcus aureus there are no growth on the nutrient gelatine and the
surface of the nutrient broth. The nutrient cultures shows the different
characteristics of the bacteria. As for broth culture, shows that the
microorganism grows in liquid medium. The slant culture shows that the
microorganisms to maintain for long term storage. The stab culture is to study
the lateral growth which is the motility characteristics of the microorganism
and plate culture is to observe the growth of microorganisms on the surface of
solid medium.
