TEST FOR SALIVARY AMYLASE

Objective: To test the presence of Amylase enzyme in the human saliva.


Materials and Solutions required: 2 Plastic Cups (14 oz), Medicine Dropper, Mashed
Potato, Iodine solution, Human Saliva.
Procedure:
1. Write the samples: In first plastic cup, please write “starch solution with saliva”
while the second plastic cup, please write “starch solution without saliva”. (For
comparison of data)
2. Add mashed potato pieces in the plastic cups and add warm water. Dissolved well.
This will served as a starch solution.
3. Add few drops of iodine solution to the liquid in the plastic cups.
4. Spit a few drops of saliva in the 1st plastic cup.
5. Notice the bluish black (dark blue) colour in the test tube.
6. Leave the sample undisturbed for 5 minutes. Observe the colour change in the
sample solution.
7. The solution gradually becomes colourless which confirms the presence of amylase
in the human saliva.
