1. Sahli Method
Principle
Hb +HCl Hematin hydrochloride The color is matched

 Procedure
1- Put drops of 0.1 N HCl in the sahli graduated tube to the mark 2.
2- Draw the blood to the 0.02ml mark using the sahli blood pipette.
3- Add the blood to the dilute acid and Mix the blood and acid.
4- Place the tube into the tube holder of the colored scale.
5- add distilled water drop by drop with stirring the solution using glass
rod.
6- Keep adding the distilled water until the color of the solution matches
the color of the glass standard.
7- Record the value of Hb content.
