MATERIALES Y MÉTODOS Las semillas de altramuz cv. Marta y soja cv. Williams se esterilizaron con
hipoclorito sódico (10%, v/v) durante 30 min y posteriormente se lavaron varias veces con agua
destilada. La germinación se llevó a cabo en oscuridad (28ºC; 3 días). Producida la nascencia, se
colocaron sobre unos recipientes de cultivo con capacidad para 3 L de disolución nutritiva y bajo
oxigenación continua. El experimento se llevó a cabo en cámara de cultivo bajo condiciones
ambientales controladas: Temperatura: 20-25ºC; fotoperiodo: 11-13h; humedad relativa: 40-60%;
intensidad luminosa: 520 µmol m-2 s -1 . El sistema de cultivo fue hidroponía sin sustrato. La
composición de la disolución nutritiva control fue la siguiente (mM): 1,5 Ca(NO3)2; 4,0 KNO3; 1,5
KH2PO4; 1,0 MgSO4. Los micronutrientes se suministraron como (µM): 36 Fe-EDDHA; 33
MnSO4..H2O; 1,6 ZnSO4.7H2O; 0,1 (NH4)6Mo7O24.4H2O; 46 H3BO3 y 1,6 CuSO4.5H2O (pH 5-
5,5). La disolución nutritiva se renovó semanalmente y cada dos días se reponía con agua el
volumen inicial. Las dosis de Cu utilizadas fueron 1,6 (control) y 24 µM (CuSO4.5H2O).El diseño
experimental correspondió a un sistema de bloques completos al azar. Se realizaron cuatro
replicados por dosis de Cu y en cada recipiente de cultivo se pusieron 3 plantas. A los 35 días de
exposición a los tratamientos de Cu, se llevó a cabo el muestreo del material vegetal. Para ello, las
plantas de altramuz y soja se dividieron en raíz y vástago y éste último a su vez en hojas y tallos y
se estimaron los pesos frescos de cada órgano. A continuación, las muestras se introdujeron en N2
líquido y se almacenaron en un congelador (MDF-U72V SANYO) a -78ºC. Posteriormente, se realizó
un fraccionamiento subcelular mediante centrifugación diferencial siguiendo las metodologías
propuestas por Hernández y col. (1998) y Wu y col. (2005). La concentración de Cu en la fracción
soluble, pared celular y orgánulos se determinó mediante espectrofotometría de absorción
atómica. Además, en muestras “in vivo” de hojas y raíces se llevó a cabo el microanálisis de
elementos mediante microscopía electrónica de barrido a baja temperatura y difracción de rayos X
(LTSEM-EDXMA).
