INSTALLATION

Automatic analyser must be installed in closed environment, adequately protected from


atmospheric agents, on a predisposed plan, of the opportune dimensions to allow an easy
use.
In case of insufficient illumination, it's necessary to provide with the installation of a
system of illumination appropriated, in order to make the user able to act under normal
light conditions.

MAROCHE s.r.l. does not charge responsibilities for the use of the analyser inside and
umproper environment.

Always leave an appropriate free space around the instrument, to


allow a proper air flow and space for final user.

For the instrument installation, arrange a point of lacing to the electric grid with a ground
connection and protected from short-circuits, as told by safety normative anticipated in
subject; the characteristics of instrument electric supply are pointed out in the electric
schemes.

The instrument is equipped of electric plant as dictated by the law 46/90 in safety law.

INSTALLATION PLACE REQUIREMENTS

1. Choose a position for the instrument that has to be far from direct sun exposition
and dust.
2. Temperature should be inside 15-30 °C range.
3. Support desk must be flat, with no vibrations, and able to support a >40 kg weight.
4. Power supply system’s requirements are: 110-240 V A.C.
5. Power consumption: 450 Watt.

INSTALLATION PROCEDURES

Unpacking and Checking

After you have unpacked your ATOM, check every accessory and ensure that the
instrument has not broken parts or dents.

Put the instrument upon a flat plan.

ATTENTION:

Before turn ON your instrument, remove the blocking screw on the back (Fig. 2)

Now you can go on with cables and wastes bottle installation (Fig.3)

DECONTAMINATION

When an instrument must be assisted or repacked, it needs first to be decontaminated.

Necessary material:

 Solution at 70 % of alcohol

 Cleaning Cloth

Procedure :

1. Clean all the external instrument surfaces with alcohol solution.

2. Clean all the inside parts that could have been in contact with hazardous material.

SOFTWARE INSTALLATION
NB.: Be sure that in your operating system the decimal separator is “.” dot
instead of “,” coma (Check it from Windows Control panel -> Language and International
Settings)

- You will find an Installation CD with every instrument


- Insert Installation CD inside you CD/DVD rom drive
- Windows will show you as follows (if auto-run is supported and activated, otherwise
provide to open it manually from My computer)

- ATOM.MSI (software installer)


- USB_RS232_DRIVER (USB/serial adapter Windows driver)
- XMET.zip (zipped folder with methods inside, to be extracted on C:\)
- file 968XXXX.modatom (where 968XXXX is your ATOM serial number)

Start now installation:

1. Copy the zipped folder called “XMET” on Drive “C:\” (root directory)
2. At the end of copy, use WinZip or WinRAR (you find WinZip and WinRAR installer
inside your Installation CD) to decompress this folder using the option “Extract to
here” (WinZip) or “Extract Here” (WinRAR).

3. After extraction is complete you will have a normal folder called XMET on C:\
4. Now start software installation double-clicking on ATOM.msi file
5. Click on “NEXT” button in every page till the end of installation

After installation is done you can start the program making double click on this icon from
your Desktop:

Main page will appear:

Click on “SERVICE” button, then log in with password “zeus” to enter SERVICE menù.

3. Search CD directory and select the file that is named with serial number of the
instrument (ex.: 9680057.modatom), then click on “OPEN” button. If everything
went right, a “LOADED” message will appear. This file (968XXXX.modatom) contains
all the necessary mechanical settings for the device. Without this, instrument will be
damaged when used.

Now you're finally able to start instrument.

NOTE: Make sure that PC is equipped with speakers, because software does produce
sound signals.

LOAD TIP POSITION

For adjust tip loading positions you have to:

- Put a disposable-tip on ATOM tray in the position that you want check (for example
1st position, immediately near S1 position)
- Select on screen it’s position (1° , from SAMPLE TIP group on left top corner)
- Adjust values for X and Y axes
- Click on TRY for to visually check if it is correctly centered
- Push SAVE TO HARD DISK after every change
- When correct position is found, click on LOAD EEPROM button
- Repeat for all the positions

On left side you have 16 positions for SAMPLE TIPS, on right side you have 16 positions for
REAGENT TIP. Follow the same procedure for both groups.
When all the positions are correctly centred, you can check the strength used by Z axes
while takes a tip. You can increase or decrease it increasing or decreasing value of OFFSET
TIP DESCEND, the more it will go down, the more strength it will use when take the tip.
Choose the most suitable for your instrument. After every value change click on SAVE TO
HARD DISK button. When you have found correct value, click on LOAD EEPROM button.

With OFFSET BLOCK X1, X2, Y and Z POSITION you can adjust position of TIP HOLDER into
hole for TRAY LOADING (allow to open and close tray and shacking movements).

With LEVEL SENSOR YES or NO options you can choose to use or not level sensor. Level
sensor allow you to work directly from samples primary tubes, but it will require use of
disposable carbon type tips (conductive). If you choose to not use level sensor you will not
be able to work directly from primary tubes and it will be necessary to pipette exact
amount of sample into sample holder vial (common 5ml tubes). In this case you will use
normal disposable tips.

SAMPLE POSITION

For adjust sample positions you have to (first load a tip from LOAD TIP POS. page):

- Put a 5ml vial into S1 position on tray


- Select on screen it’s position (1°, from SAMPLE POSITION group)
- Press TRY button
- Check that position is correctly centered inside vial
- If not, adjust X and Y value
- Push SAVE TO HARD DISK after every change
- When correct position is found, click on LOAD EEPROM button

In order to check if height of tip is correct, you have to:

- Click on TRY button


- When tip is inside vial, block vial with your fingers and be sure that you can move up
and down tip holder and vial at the same time, without moving them one respect the
other
- Turn OFF ATOM
- Move up Z axes up and check if tip extremity is inside lower side of vial (under the
cut, that indicates more or less volume occupied by dead volume), being sure that it
not touch vial bottom
- If you need you can adjust this value with B_CUV DESCEND

In case you use LEVEL SENSOR, height will be adjusted with SENSOR OVER DESCEND value.

REAGENT POSITION

To adjust reagent positions you have to (first load a tip from LOAD TIP POS. page):

- Put a 5ml vial into R1 position on tray


- Select on screen it’s position (1°, from REAGENT POSITION group)
- Press TRY button
- Check that position is correctly centered inside vial
- If not, adjust X and Y value
- Push SAVE TO HARD DISK after every change
- When correct position is found, click on LOAD EEPROM button

In order to check if height of tip is correct, you have to:

- Click on TRY button

UNLOAD TIP POSITION

To adjust unload tip position you have to:

- Load a tip from LOAD TIP POS. page


- Click on TRY button
- Check if instrument correctly unload the tip
- If not, you can adjust movement changing OFFSETs values for X, Y and Z axes
- Push SAVE TO HARD DISK after every change
- When correct position is found, click on LOAD EEPROM button

WELL DISPENSATION

To adjust positions for pipetting you have to (first load a tip from LOAD TIP POS. page):

- Put an ELISA micro-plate on tray with at least of strip of 8 wells


- Select the position that you want to check (1st position in our example pic)
- Press TRY button
- Check if tip extremity is correctly centered on each wells (check all 96 positions)
- If not, you can adjust movement changing values for X and Y axes
- Push SAVE TO HARD DISK after every change
- When correct position is found, click on LOAD EEPROM button

With OFFSET TIP DESCEND value you can adjust height of tip extremity upon the wells. The
most suitable distance is more or less 2 mm from well’s top.

WASH POSITION

To adjust positions for washing you have to:

- Put an ELISA micro-plate on tray with at least of strip of 8 wells


- Select the position that you want to check (1st position in our example pic)
- Press TRY button
- Check if washer’s needles are correctly centered on each wells (check all 96
positions)
- If not, you can adjust movement changing OFFSETs values for X and Y axes
- Push SAVE TO HARD DISK after every change
- When correct position is found, click on LOAD EEPROM button

To check needles height for washing procedure you have to hear them lightly touch well’s
bottom, after you have pressed TRY button.

If you click on TRY FULL, you will see needles go inside the well (the one you choose from
PLATE-DISP. group of 96 positions, 1° in our example pic) than go up again and stand
immediately above well’s top. In this way you can see position from which instrument is
going to dispense wash buffer inside the well during washing procedure. To adjust this
height you can change WASHING LIFT UP value.

If you click on TRY WASH, instrument will perform a wash step to let you check that
washing is correctly executed.

From this page you can check if pumps are working, clicking ON or OFF on WASTE PUMP.
This will turn ON and turn OFF ATOM pumps.

From this page you can check as well electro-valve functioning, pressing in sequence V1
ON, V1 OFF, V2 ON, V2 OFF, etc. to see if they are working or not. Usually when turn
ON/OFF an electro-valve you can hear a “snap”, to indicates it’s correct functioning.

With HYDRAULIC FILLING you can adjust quantity of water to be used during tubing filling,
we suggest you to leave those values as they are, because they are made by factory. This
values are strictly related to length of hydraulic tubes, starting from the one from WASH
BUFFER bottle, till the ones inside your ATOM. For this reason, if you will need to replace
one or more hydraulic tubes we warm ask you to pay attention and replace only with new
tubes with same diameter and same length.

With INCUBATION COVER POSITION you can adjust position of black cover over the
humidity chamber upon the incubator, in this way you can let it stay in the most centered
position to cover at the best the micro-plate during incubation time.

Finally, in the bottom left corner you find WASH FILLING button. Clicking on it, instrument
will perform a cycle of wash filling. This could be useful for service staff and technical
people when they want to manually perform instrument cleaning.

READ POSITION

To adjust reading positions you have to:

- Put aluminium micro-plate provided with your ATOM (on its left side you will find a
little cut for each strip of 8 wells) on ATOM tray
- Click on 1st position on PLATE-READ
- Click on TRY button
- Check that cut for first strip of 8 on micro-plate (A1 to A8) is aligned with first cut on
OPTIC BLOCK, relative to 450nm filters (on optic block there are 4 cuts, first and
second - towards the internal part of ATOM - are for 450nm filters, third and fourth -
towards the external part of ATOM - are for 620nm filters)

In order to check if ATOM is going to perform reading step in a correct way, we have to

clearly understand how ATOM reading works:

ATOM optics count 16 independent channels, 8 for 450nm and 8 for 620nm. Referring
to OPTICAL CHANNELS buttons from above pic, you can identify buttons from 1 to 8 as
450nm filters and buttons from 9 to 16 as 620nm filters. Filters are not in the same
row, but shifted 4 by 4 for space reason (in this way optic block is not too wide). So,
450nm filters will be divided in this way:

- First row: channels 1, 3, 5 and 7 (450nm)


- Second row: channels 2, 4, 6 and 8 (450nm)

And 620nm filters will be divide in this way:

- Third row: channels 9, 11, 13 and 15 (620nm)


- Fourth row: channels 10, 12, 14 and 16 (620nm)

When ATOM start to read at 450nm, it will follows this scheme:

Channels 1, 3, 5 and 7 from first optic row will read wells A1, A3, A5 and A7 from first strip
of 8 well, then all the tray will shift further to the second row of channels: 2, 4, 6 and 8 will
now read A2, A4, A6 and A8 wells to complete first strip of 8 well. Then ATOM tray will
shift further to let second strip be read: B1, B3, B5 and B7 will be read from channels 1, 3,
5 and 7 (450nm), B2, B4, B6 and B8 will be read from channels 2, 4, 6 and 8 (450nm), and
so on.
For this reason you need to align every strip of 8 to 2 different row of channels (for each
wavelength). You can do that only adjusting X OFFSET value (as usual, click on SAVE TO
HARD DISK button after every change, before push TRY button). And because every strip of
8 is on the same line on ELISA micro-plate you can adjust only the first well position to
have the entire strip of 8 wells adjusted. This means that you actually have to adjust only
position for 1°, 2°, 9°, 10°, 17°, 18° …and so on.

At the end, if you need to adjust these values for 620nm as well, all you have to do is
adjust OFFSET 620 value, when done, all the positions for 620nm filters will be
automatically set.

In this page you have also a REFERENCES box, in which software will show you different
numbers, depending on what we ask it to do. For example, when you are in READ
POSITION page, if you press RESET button on ATOM (just in front of it, near the blue status
LED) and wait till the end of reset movements, software will show you the value “99” on
REFERENCES box. This only means that communication between ATOM and PC is OK.

You can use it as a tool for checking data transmission immediately after software
installation.
REFERENCES box have another purpose as well: it show you lamp value for each channel. If
you click on 1 on OPTICAL CHANNELS group and then on SINGLE READ button, it will show
you lamp value. Normal blank value for each channel is more or less 28000 (+/- 1000).
Normal dark value is more or less 2000 (+/- 200).
If you click on LOOP READ it will show you a REAL TIME value for the selected channel.
When you want to switch to another channel, you have first to press RESET button on
ATOM. LOOP READ is useful when you need to adjust value for a specific lamp, you can
increase or decrease each lamp value by its related trimmer on pre-amplification board (16
blue trimmers are present on it) and you will see value change in real time in REFERENCES
box.

NOTE: in order to check dark values, ATOM tray must be CLOSED.

From this page you can also switch between 25 °C (r.t.) and 37 °C, clicking on
TEMPERATURE 25 RES=1 button or 37 RES=0 button. In this way you can easily understand
if Peltier system is working, touching incubator to feel changing in temperature.

We suggest you to not use FULL/HALF POWER command, it is for factory checking only.
You will never need to use it for maintenance.

SYRINGE VOLUME

To adjust pipetting volume you simply need to follow indications on screen (first load a tip
from LOAD TIP POS. page):

- Put one well in A1 and one well in A2


- Put a 5ml vial in R1 position with an amount of bi-distilled water as indicated in PUT
INTO THE PLATE 8 WELLS in 1° LINE...etc
- Close ATOM tray
- Select the volume you want to check (5ul, 10ul, ...)
- Press TRY button

Instrument will dispense the selected amount inside the well, at this point you can aspire it

with an high quality manual pipette to make comparison. If volume is not correct, you can
adjust value for that volume changing OFFSET POSITION. As usual, after every change you
must press SAVE TO HARD DISK button. At the end, when all the volume are correctly
selected, you can press LOAD EEPROM button before leave the page.

PRINT PARAMETER

From this page (as seen in Installation procedure) you can LOAD mechanical settings, SAVE
mechanical settings (on a new file, or upgrading the old one) and PRINT mechanical
settings on paper.

You can LOAD mechanical settings from ATOM CD software, every ATOM has its own CD
with its S/N written on it in order to avoid any mistake. You can also SAVE for changes on a
new file on your PC or on an external HARD DRIVE/USB PENDRIVE.

PROGRAMMED MAINTENANCE

Maintenance has to be performed when the device is not active, to avoid any risk or
danger for technical staff, and has to be performed by authorized and qualified personnel
only.
A periodic control of device is expected every 6 months to verify efficiency, performance
and correct working.

Daily Maintenance

 Remove ELISA micro-plate, empty WASTE bottle, empty and clean WASH BUFFER
bottle, empty TIP WASTE closet, perform an hydraulic filling.
 Shutdown with washing solution.

Semestral Maintenance

 Clean and lubricate mechanic moving parts


 Check lamp values
 Clean electrovalve
 Clean tip holder
 Clean washer needles

Annual Maintenance

 Replace tips holder (if necessary)


 Replace silicone tubes (hydraulic tubes)
 Check photometry
 Check belts tensions
