The DNA ration at 260/280=1.8-2.0 shows the purity of DNA samples which is required for
restriction digestion experiment. The sample No 1, 2, 7, 8 and 11 showed less than1.8 OD ratio
which indicates the DNA is not pure. Therefore, these samples revealed proteins contamination
that cannot generate proper restriction digestion pattern as DNA itself when use. Ergo, these
DNA samples cannot be used for restriction digestion experiment.

The volume will be : 10 x 25 x 0.5 = 125 cc
Therefore, 125 ml of the 1.25% agarose gel is to be used.
Total amount of TAE used is 125 ml.
Total amount of agar used is 1.56 gram because you need 1.25 grams of agar per 100 ml
of TAE. Therefore, by ratio and proportion, (1.25 g x 125 mL)/100 mL.

b. There must be an error during the electrophoresis. It seems that in the photo, the
visualization was improper. It seems that the material use in visualizing the samples were
too high, whether it was a dye or a radioactive probe.
c. To resolve this, the controlled and correct process should be followed to create a
proper visualization. The exact amount to use for the process should not be too high or
too low to create a visual that is accurate and is precise.
5. A. If child 3,4, and 6 have the disease, this means that the marker for the disease allele is
a single band. Therefore, other who have extra bands do not have the disease. Meaning, it
could be a recessive disorder.
B. The carriers will be the people with this single band plus another band which is 1, 2, 8,
9, 10, and 11.
b. Smith daughter is not a carrier. To be a carrier, she should have the band and an additional
band.
c. This is if Dad Jones is affected too.
