CALDOB

(Calcium Dobesilate 500 mg)

Mechanism of Action
• Decreases platelet aggregation by inhibiting platelet aggregation factor (PAF).
This prevents formation of thrombus.
• Decreases hyper-viscosity of blood by inhibiting lipid peroxidation of
erythrocytes. This increases RBC membrane flexibility and helps in free flow
of blood, enhances release of tissue plasminogen activator (TPA), thus
increases fibrinolytic activity.
• Strengthens blood vessels, improves integrity of blood vessels.
• Reduces hyper-permeability by synthesizing nitric oxide which helps to relax
blood vessels, reduces oedema, heaviness and pain in legs.
• Also inhibits synthesis of prostaglandins.

ROLE OF CALDOB IN VARICOSE VEIN


What is a varicose vein?
Dilation of veins in lower limbs is known as varicose veins. Varicose veins are
inflamed, twisted and swollen veins. Major symptoms of varicose veins: oedema,
heaviness in the leg, paresthesia, day and night cramps.

• CALDOB reduces symptoms of varicose veins by inhibiting thrombosis, thus


improves the circulation.
• CALDOB reduces blood pressure in pregnant women.
• CALDOB also decreases capillary hyper-permeability.
• CALDOB acts on endothelial layer of blood vessels and increases synthesis of
nitric oxide, relaxes the blood vessels and close gaps.

ROLE OF CALDOB IN DUB


Vasoprotector: It normalizes capillary hyper-permeability and reduces capillary
fragility
CALDOB effectively reduces DUB by acting on various prostaglandins, responsible
for uterine bleeding.

ROLE OF CALDOB IN HAEMORRHOIDS


• CALDOB is the most effective treatment in haemorrhoids.
• CALDOB corrects the root cause, i.e. veinous swelling and thrombosis. So
excellent and faster result in both fisrt and second degree of haemorrhoids.
• CALDOB reduces veinous swelling by decresing capillary hyper-permeability
and also inhibits thrombosus in th eveinous, plexus to its anti-platelet and
fibrinolytic proeprties.

CALDOB causes erection in penis by causing synthesis of nitric oxide and acts
specifically on penile arteries.

TREATMENT IN ED
1. Phosphodiesterase enzyme inhibitors (pde-5 inhibitors). Exp: sildnafil,
tadalafil.
2. Nitric oxide donors. Exp: calcium dobesilate (CALDOB).

SILDENAFIL: Mechanism of action


• Acts by blocking the action of phosphodiesterases. It firther inhibits the
degradation of cGMP (cyclic guanosine monophosphate).
• cGMP causes the smooth muscles of the arteries of the penis to relax corpus
carvernosum (sponge like tissues of penis) to fill with blood.

Limitation of PDE-5 Inhibitors (sildenafil)


• Sudden hair loss.
• Ringing in the ears.
• Reddening of the face and neck (flushing).
• Dizziness.
• Headache.
• Conte indicated with nitrate drugs (used in pain treatment).
• Contraindicated with alpha blockers (used to treat high B.P. and be nigh
prostatic hyperplasis (BPH).

Mechanism of action of CALDOB in E.D.


• Endothelium derived hyperpolarizing factor (EDHF) plays important role in
the endothelium dependent relaxation of human penile resistant arteries
(HPRA).
• Nitric oxide (NO) is a key mediator of endothelium dependent relaxation.
• Calcium dobesilate (CALDOB) potentiates endothelium dependent relaxation
of HPRA by enhancing the production or the action of EDHF.

TESTIMONIALS
• Phsophodiestrases-5 (PDE-5) inhibitors are less effective in the treatment of
E.D. in diabetic men.
• Unlike sildenafil, CALDOB fully reverse diabetes induced endothelium
dysfunction in penile resistance arteries.

DOSAGE: one cap BID for 4 to 8 weeks or till the satisfactory result.
