seutellarioides) EXTRACT AS AN ALTERNATIVE pH


INDICATOR TEST STRIP IN THE URINE SPECIMEN

INTRODUCTION
Coleus plant (Plectranthus seutellarioides)
also known as mayana or lampunaya plant
in the Philippines.

It comes in variety of colors like yellow, red,


pink, green and purple.

Coleus plant is rich in anthocyanin because


of its color.
INTRODUCTION
We are going to observe the
effectiveness of coleus plant extract
as a pH indicator strip in urine
specimens.

 Coleus plant extract will be use in


making pH indicator solution.
OBJECTIVES
General objectives:
This study will tend to determine the
effectiveness of coleus (Plectranthus
seutellarioides) extract as a pH indicator
test strip in various urine specimens.
OBJECTIVES
SPECIFIC OBJECTIVES:
Determine the acidity or alkalinity of
various urine specimens and its
corresponding condition, using coleus
(Plectranthus seutellarioides) extract as pH
indicator.
Determine the effectiveness of anthocyanin
as an alternative for acid-base testing.

HYPOTHESIS OF THE STUDY

There is no significant effect on the coleus


(Plectranthus seutellarioides) extract as a
pH indicator test strip in various urine
specimens.

This study will be deemed important to
the following people, group &

Pharmaceutical companies - This


may give insightsand potential
pharmaceutical ingredients that give
various beneficial health effects.

concerning homeostatic regulation
of the pH in the body’s
extracellular fluid.

acid-base homeostasis by testing urine sample.

SCOPE AND LIMITATION
This study will focus on the effectiveness of coleus
(Plectranthus seutellarioides) extract as a pH indicator test
strip in the urine specimen. The researcher will observe if
coleus (Plectranthus seutellarioides) can use as a pH
indicator strip after doing some experiments and test. This
result will help the researchers determine if coleus
(Plectranthus seutellarioides) would make a successful pH
indicator.
SCOPE AND LIMITATION
All investigatory activities and
laboratory works will be conducted at
science laboratory at Riverside College
Inc., On February 2019.

This study is an experimental research
emphasizing on the effectiveness of coleus’
anthocyanin property on indicating urine’s
ph.

Collection and preparation of the extract and
solution
The researchers will be collecting coleus
leaf.
In the preparation of coleus leaf indicator
solution, 1kg of coleus leaf, put it in a
blender and add 500ml of boiling water.
Blend the water and coleus leaves.

 Keep the mixture blending until the water is
purple.
Transfer the mixture into a pan (choose a
container that is stain resistant) and pour the
mixture through a strainer to remove any pieces
of leaf from the indicator solution.

Add 50mL of isopropyl alcohol to the solution to
protect from bacterial growth.
 The alcohol may alter the solution, if this
happens, add vinegar until the solution goes
back to red purple.

Prepare an acid free paper and then soak it into
the solution.
 Allow paper to air dry on a towel leave it
overnight. Cut the paper into strips.
Put strips inside a dry airtight container and
place it in a cool area.

The pH indicator strip will be used to
indicate the urine’s ph.
Collect the specimen (urine) needed in a
test tube up to its neckline.

Procedure:
Test strip will be soaked for 1 second in
the urine specimen.
For a few seconds observe for the
change color in the strip. Each color has
a corresponding pH result.
