2. Inhibarea radicalului anion superoxid.
Anionul superoxid are o toxicitate mare i este
generat n organismul uman ca msur defensiv
mpotriva tulpinilor patogene. Este duntor atunci cnd
rezult n urma activitii mitocondriale. S-a utilizat
metoda prezentat de Bhatt i colab., 2013 (Creative
Commons Attribution License), cu mici modificri:
- 0.5 mL nitro tetrazoliu albastru (NBT) 0.3 mM;
- 0.5 mL nicotinamid adenin dinucleotid
(NADH) 0.936 mM;
- 1 mL prob;
- 1 mL tampon Tris-HCl 16 mM, pH 8;
- 0.5 mL fenazin metosulfat (PMS) 0.12 mM;
- n paralel se realizeaz un martor, n care nu se
adaug prob;
- Se raporteaz rezultatele fa de un standard, de
exemplu acid ascorbic 1 mg/mL;
- Eprubetele se pstreaz 5 minute la 250C;
- Se citete absorbana la 560 nm cu un
spectrofotometru Helios ;

- Se calculeaz inhibarea SO (%) = 100.

3. Determinarea puterii de reducere.
n aceast metod compuii bioactivi ce pot dona
un electron determin reducerea Fe3+ n Fe2+. Rezultatele
obinute reprezint un indicator direct al capacitii
antioxidante a unui produs. S-a utilizat metoda
prezentat de Bhatti i colab., 2015 (Creative Commons
Attribution License), cu mici modificri:
- 0.2 mL prob;
- 0.5 mL tampon fosfat 0.2M, pH 6.6;
- 0.5 mL fericianur de K 1%;
- Incubare 20 minute la 500C ntr-un termostat
Memmert;
- 0.5 mL acid tricloracetic 10%;
- n alte eprubete se pun urmtoarele:
o 0.5 mL amestec iniial fr sediment;
o 0.5 mL ap distilat;
o 0.1 mL FeCl3 0.1%;
o Se raporteaz rezultatele fa de un
standard, de exemplu acid ascorbic 1
mg/mL;
o Se citete absorbana la 700 nm cu un
spectrofotometru Helios ;
o Rezultatele se interpreteaz n funcie de
valoarea citit la spectrofotometru.

5. Inhibarea peroxidrii lipidelor
Metoda, n toate variantele sale, este recunoscut
ca fiind adevratul indicator al activitii antioxidante a
unui produs. S-a utilizat metoda prezentat de Abdullah
i colab., 2012 (Creative Commons Attribution License),
cu mici modificri:
- 0.1 mL prob;
- 1 mL emulsie din glbenu de ou n tampon
fosfat0.1M pH 7.4;
- 0.1 mL FeCl2 1 mM;
- Eprubeta se introduce o or pe baie de ap la
370C;
- 0.5 mL acid tricloracetic 15%;
- 1 mL acid tiobarbituric n acid acetic;
- Eprubeta se incubeaz 10 minute la 1000C;
- Se raporteaz rezultatele fa de un standard, de
exemplu acid ascorbic 1 mg/mL;
- n paralel se realizeaz un martor n care nu se
pune prob;
- Se citete absorbana la 532 nm cu un
spectrofotometru Helios ;
- Se calculeaz Capacitatea inhibitorie (%) =
