 Introducere
ICP-MS este folosită ca metodă de analiză
multielement a probelor clinice încă din
anii 1980 , fiind utilă în chimia analitică
datorită flexibilității, sensibilității și
reproductibilității.

 Scopul studiului
Scopul studiului a fost dezvoltarea unei metode analitice pentru
determinarea concentrației de nichel (Ni), arsen (As), cadmiu (Cd) și
plumb (Pb) în probe de păr prin spectrometria de masă cu plasmă
cuplată inductiv (ICP-MS).
 Metoda studiului
Părul uman de la diferite grupe de vârstă a fost colectat din Beijing, China. Probele de păr au fost
obținute de la persoane care nu aveau părul vopsit sau tratat. Deoarece bărbații aveau părul scurt, părul
era tuns din toate zonele scalpului. Pentru femelele cu păr lung, doar porțiunea distală a părului a fost
tunsă. Probele de păr au fost păstrate în pungi de polietilenă la temperatura camerei până la analiză.
Mostrele de păr au fost tăiate în bucăți cât mai mici pe cât posibil și au fost supuse ultrasonarii de
cinci ori cu 25 ml acetonă-apă-apă-apă-acetonă [10], timp de 10 minute de fiecare dată și apoi uscate
într-un cuptor peste noapte la 60°C. Scopul spălării părului este de a elimina orice ulei și material gras
de la suprafața părului [12].
O portiune de 0,5 g din proba preparată a fost cântărită cu precizie utilizând o balanță analitică cu patru
zecimale și transferată într-un vas de digestie din politetrafluoretilenă cu capac filetat. Un amestec de 2
ml de acid azotic și 1 ml de peroxid de hidrogen 30% a fost adăugat la temperatura camerei și lăsat să se
predigere timp de 12 ore. Vasele au fost apoi sigilate și introduse în cuptorul la 160 °C timp de 4
ore. Odată ce digestia a fost completă, vasele au fost lăsate să se răcească la temperatura
camerei. Digeratul a fost diluat la 10 ml cu apă ultrapura și in final, analizat prin ICP-MS.
 Metoda studiului
ICP-MS este echipat cu un cuarț concentric Meinhard nebulizator, cameră de pulverizare
ciclonică, prelevator de nichel și con skimmer. robele martor de reactiv au fost digerate în același
mod ca și pentru mostrele de păr și au fost utilizate pentru a corecta citirile instrumentului.
Părul uman de la diferite grupe de vârstă a fost colectat din Beijing, China. Metalele în cauză -
Cd, Cr, Pb, As și Hg - au fost analizate, iar nivelurile de metale în raport cu vârsta, sexul și aportul
alimentar au fost evaluate în continuare. Rezultatele au arătat că cel mai înalt nivel al metalelor a
fost prezentat de Pb, cu o concentrație medie de 1,557 ± 0,779 mg/kg, urmat de Cr (0,782 ± 0,394),
Hg (0,284 ± 0,094), As (0,127 ± 0,078) și Cd (0,071 ± 0,032), după o ordine descrescătoare a Pb >
Cr > Hg > As > Cd. Concentrațiile de metale grele au variat foarte mult între diferitele grupe de
vârstă, iar concentrațiile mai mari de Cd, Cr, Pb și As au apărut în părul feminin, în timp ce
concentrații mai mari de Hg au fost găsite în părul masculin.
 Asigurarea și controlul calității

Probele martor de reactiv au fost digerate în același mod

ca și mostrele de păr și au fost utilizate pentru a corecta
citirile instrumentului. Pentru a valida metoda de
determinare a concentrației de metale, s-a folosit materialul
de referință pentru părul uman din SUA.

 Rezultate si discutii

Plumbul (Pb) reprezintă cea mai mare concentrație medie în

comparație cu celelalte elemente din eșantioane unde media
și standardul de deviatie al concentratiei de Pb în probe este
de 75,07 ± 37,65 μg/g. Pb este un element toxic chiar și în
concentrație mică în corpul uman și una dintre cele mai
populare analize de metale grele din cauza numeroaselor
efectele adverse asupra sănătății. In cantitati toxice poate
inhiba transportul de oxigen și calciu și modifica transmisiile
nervoase în creier. Abaterea standard a concentratiei de
nichel este de 21,71 ± 20,39 μg/g.
 Concluzii
În acest studiu o metodă de analiză cantitativă pentru determinarea a unor
metale (Ni, As, Cd și Pb) în zece probe de păr prin ICP-MS a fost dezvoltat și
validat. Rezultatul oferă suficiente dovezi că se poate concluziona că această
metodă este valabilă pentru utilizare în aspect toxicologic în ordine pentru a
determina nivelul cantitativ al toxicului și acut al metalelor grele în mostre de păr.
 Concluzii
În concluzie, în prezentul studiu au fost analizate concentrațiile de
metale grele de Cd, Cr, Pb, Hg și As din probe de păr prin ICP-MS.
Studiul oferă suficiente dovezi care pot concluziona că această metodă
este valabilă pentru utilizare în aspect toxicologic pentru a determina
nivelul cantitativ al metalelor grele în mostre de păr.
