nucleases - ZFNs) combină un domeniu de legare ADN “deget de zinc”
cu enzima (nickase) FokI ce cauzează ruperi monocatenare. Fiecare
“deget de zinc” recunoaște o tripletă de baze și, în mod obișnuit, în fiecare
domeniu țintă sunt prezente trei până la șase degete de zinc. Clivarea se
produce înafara secvenței țintă și necesită o pereche de nucleaze “degete
de zinc” (ZFNs), fiecare legându-se la o catenă a ADN. (B) In mod
similar nucleazelor “deget de zinc”, nucleazele efector similare
activatorilor transcripției (transcription activator-like effector nucleases -
TALEN), au două domenii: un domeniu de legare ADN și un domeniu
pentru enzima FokI. Domeniul de țintire este compus din repetiții de 33-
35 de aminoacizi, fiecare legându-se la o singură nucleotidă. Mecanismul
de clivare a nucleazelor efector similare activatorilor transcripției
(TALENs) este identic celui al nucleazelor deget de zinc (ZFNs).
(C) Repetițiile palindromice scurte intercalate regulat (clustered regularly
interspaced short palindromic repeats - CRISPR) este un sistem de
